The subclass 186 has three streams, all for skilled workers nominated by an employer to live and work in Australia permanently.
The Direct Entry stream is for applicants who have not worked in Australia (or only for a short time) and who meet the skills and experience requirements directly. The Temporary Residence Transition stream is for people who have worked for their nominating employer on an eligible temporary skilled visa. The Labour Agreement stream is for workers nominated by an employer that has a labour agreement with the Australian Government.
The right stream depends mainly on your work history in Australia and your employer's sponsorship arrangements.
